dc.description.abstractIspitivanjem osetljivosti jedinki domaćeg miša izlovljenih iz divljih populacija sa sedam lokaliteta, primenom metode testa hranjenja u trajanju od 21 dan, utvrđena je rezistentnost na bromadiolon (0,005 %) na dva lokaliteta. Takođe, na jednom od ostalih lokaliteta utvrđen je znatno smanjen nivo osetljivosti na ovaj antikoagulant. Primenom istog eksperimentalnog dizajna na potomke rezistentnih jedinki utvrđena je rezistentnost kod 38,46 % jedinki. Manji procenat smrtnosti i duži period preživljavanja kod osetljivih jedinki utvrđen je kod ženki. Takođe, manja promena telesne mase utvrđena je kod osetljivih i rezistentnih ženki. Kod ženki je utvrđena veća letalna i neletalna doza u odnosu na mužjake u svim sprovedenim testovima hranjenja. Molekularnim metodama utvrđeno je prisustvo Leu128Ser i Tyr139Cys varijanti u trećem egzonu vkorc1 gena. Takođe, kod pojedinih jedinki utvrđeno je istovremeno javljanje obe varijante, odnosno kombinacija varijanti Leu128Ser/Tyr139Cys u pomenutom egzonu. VKOR varijanta Tyr139Cys i kombinacija varijanti Leu128Ser/Tyr139Cys potvrđene su i kod osetljivih i kod rezistentnih jedinki. Varijanta Leu128Ser utvrđena je samo kod osetljivih jedinki. U egzonu 2 nisu utvrđene promene u sekvenci vkorc1 gena, dok je u egzonu 1 utvrđena do sada neutvrđena promena u sekvenci ovog gena, Ala21Thr. Ova varijanta utvrđena je samo kod dve rezistentne jedinke. Kod jedne jedinke bila je u kombinaciji sa Tyr139Cys varijantom. VKOR varijanta u kombinaciji sa polom ispoljila je značajan uticaj na nivo osetljivosti jedinki na bromadiolon. Nosioci Tyr139Cys varijante samostalno ili u kombinaciji sa drugim varijantama imali su smanjen nivo osetljivosti na bromadiolon u odnosu na nosioce Leu128Ser varijante. Isto se odnosi i na nosioce nove, Ala21Thr, varijante. Smanjen nivo osetljivosti odražavao se preko dužeg vremena preživljavanja, veće konzumacije bromadiolon mamca i manjih promena u telesnoj masi. Mamci sa sadržajem difenakuma (0,005 %) ispoljili su 100 % efikasnost na jedinke domaćeg miša koje su bile rezistentne na bromadiolon u testovima hranjenja u trajanju od 21 dan. Takođe, mamci sa sadržajem brodifakuma (0,005 %) ispoljili su efikasnost od 90 % i 100 % u testovima sa kratkoročnim izlaganjima od 1 i 2 dana.sr
dc.description.abstractThe susceptibility of house mice trapped at 7 locations was examined in 21-day feeding tests which revealed bromadiolone (0.005 %) resistance in two locations. In one of the other locations, we detected a significantly lower susceptibility to that anticoagulant. Applying the same experimental design on the offspring of resistant animals, we detected resistance in 38.46 % of the animals. Considering the susceptible animals, females had lower mortality percentages and longer survival intervals. Also, females sustained less variation in body weight both among the susceptible and resistant animals. Females also had higher lethal and non-lethal doses than males in all feeding tests. In molecular testing, the Leu128Ser and Tyr139Cys variants were detected in exon 3 of the vkorc1 gene. In some of the animals, both variants were found to occur simultaneously as the Leu128Ser/Tyr139Cys combination in exon 3. The VKOR variant Tyr139Cys and the combination of variants Leu128Ser/Tyr139Cys were confirmed both in susceptible and resistant animals. The variant Leu128Ser was only found in susceptible animals. No change in the vkorc1 gene sequence was detected in exon 2, while a so far unreported change in that gene sequence, the Ala21Thr, was detected in exon 1. The variant was found in two resistant animals only. In one of these animals, it occurred in combination with the Tyr139Cys variant. VKOR variant in combination with gender was found to affect significantly the susceptibility of animals to bromadiolone. Animals carrying the Tyr139Cys variant alone or its combination with the other showed a lower level of susceptibility to bromadiolone than those carrying the Leu128Ser variant. The same applies to the carriers of the new Ala21Thr variant. Lower susceptibility was detected through longer duration of survival, greater intake of bromadiolone baits and less variation in body weight. Baits containing difenacoum (0.005 %) demonstrated 100 % efficacy against the house mice previously found to be bromadiolone-resistant in the 21-day feeding tests. Baits containing brodifacoum (0.005 %) showed 90 % and 100 % efficacy in tests with brief exposure periods of 1 and 2 days.en
