dc.description.abstractУ свету живи више од 50 милиона људи са епилепсијом. Основ лечења епилепсија представља сузбијање напада помоћу антиепилептичких лекова, али чак у приближно трећине случајева постоји фармакорезистентна епилепсија. Магнезијум је биоесенцијални минерал са вишеструким функцијама у живим системима, посебно значајан за ексцитабилна ткива. Антиконвулзивно дејство магнезијума има клиничку примену у лечењу еклампсије. Антиепилептички потенцијал магнезијума недовољно је искоришћен. У овој студији је испитивано антиепилептичко дејство Mg2+ на експерименталном моделу епилептиформне активности индуковане у Рецијусових неурона пијавице (РНП) H. sanguisuga блокадом Ca2+ канала помоћу Ni2+. Коришћене су методе класичне електрофизиологије да се испита механизам супресивног деловања Mg2+ на овом моделу несинаптичке епилептогенезе. Вршено је интрацелуларно регистровање потенцијала мембране, директна електростимулација ћелије и одређивање улазног отпора мембране (УОМ). Индукована епилептиформна активност одликовала се стварањем осцилација потенцијала мембране у облику пароксизмалних таласа деполаризације (ПТД). Мерени су параметри: ПТД фреквенција, ПТД трајање, ПТД амплитуда и број акционих потенцијала по ПТД-у, и рачунат интензитет епилептиформне активности. Нађени резултати показују да у опсегу тестираних концентрација (од 1 mmol/l до 20 mmol/l) Mg2+ супримира Ni2+ – индуковану епилептиформну активност, статистички значајно смањујући вредности свих параметара на дозно – зависан начин. dc.description.abstractThere are more than 50 million people globally living with epilepsy. The basis to epilepsy treatment is controlling epileptic seizures with antiepileptic drugs. However, in as many as around 1/3 of cases pharmacoresistance occurs. Magnesium is a bioessential mineral with multiple functions in living organisms, especially important for excitable cells. Anticonvulsive effect of magnesium is used to treat eclampsia, but its antiepileptic potential is therapeutically not used sufficiently. This study examines the antiepileptic effect of Mg2+ on an experimental model of epileptiform activity induced in Retzius nerve cells (RNC) of the leech H. sanguisuga by blocking Ca2+ channels with Ni2+. Methods of classical electrophysiology were used to investigate the mechanism of suppressive effect of Mg2+ on this model of nonsynaptic epileptogenesis. We performed intracellular membrane potential recording, direct electrostimulation of the cell and determination of the input membrane resistance (IMR). The induced epileptiform activity was characterized by generation of membrane potential oscillations in a form of paroxysmal depolarization shifts (PDSs). Several parameters were measured: PDS frequency, PDS duration, PDS amplitude and the number of action potentials per PDS. The intensity of epileptiform activity was calculated. The results obtained show that within the range of concentrations tested (from 1 mmol/l to 20 mmol/l) Mg2+ acts to suppress Ni2+ – induced epileptiform activity, causing a statistically significant decrease in the values of all parameters observed in a dose – dependent manner. Half – maximal inhibitory concentration was found to be approximately 10 mmol/l Mg2+. Concentration of 20 mmol/l Mg2+ completely abolished epileptiform activity.
