Starost i poreklo populacija vrste Salvia officinalis L. (Lamiaceae) u centralnom delu Balkanskog poluostrva - morfometrijski i molekularni pokazatelji

The age and origin of Salvia officinalis L. (Lamiaceae) populations from the central part of the Balkan peninsula - morphometrical and molecular parameters

Abstract
zahvata jadransko primorje i zaleđe. Postavlja se međutim, pitanje autohtonosti disjunktnih, kontinetalnih populacija u centralnom delu poluostrva, jer se ova termofilna vrsta od davnina koristi u lekovite svrhe, pa postoji mogućnost da su ove populacije antropogene, nastale nakon ledenog doba. Da bi se utvrdila starost i poreklo ovih disjunktnih populacja uzorkovano je 8 populacija za molekularna i 11 populacija za morfmetrijska ispitivanja. Za filogeografski deo istraživanja korišćene su sekvence dva intragenska spejsera hloroplasta 3’rps16-5’trnK and rpl32-trnL. Detektovano je sedam haplotipova koji nisu bili nasumično zastupljeni i poseduju filogeografsku strukturu, kao i visok ukupni genetički diverzitet (HT = 0.695) i genetičku diferencijaciju (GST = 0.682). Prema molekularnom satu i bajeziskoj analizi, razdvajanja predačkog genskog pula S. officinalis na dve linije desilo se u pliocenu (pre 3,267 mil. godina), što se podudara sa vremenom kada su centralni deo Balkana prekrile ve...like vodene površine i po svoj prilici podelile pretpostavljeni predački areal S. officinalis. Strukturirana, manje divergentna kontinetalno-primorska linija (CAL, Hd = 0.426, π = 0.00106) i divergentnija isključivo primorska linija (PAL, Hd = 0.403, π = 0.00257) nemaju odlike nedavne demografske ekspanzije i danas se preklapaju na jugoistočnom delu jadranskog primorja, koje je označeno kao glavni glacijalni refugijum S. officinalis. Četiri populacije iz kontinentalnog dela Balkanskog poluostrva, koje čine kontinetalnu podliniju CAL odlikuje skoro isključivo prisustvo posebnog haplotipa koji je redak u primorskoj podliniji CAL i odsutan u PAL. Zbog bliske genealoške veze ovog i ancestralnog haplotipa prema parsimonijskoj statističkoj analizi, kao i zbog specifičnih odlika kontinetalnih populacija, one su okarakterisane kao reliktne i refugijalne...

Within Balkan Peninsula, Salvia officinalis L. (Lamiaceae), common or Dalmatian sage, grows naturally along the Adriatic coast. However, the indigeneity of its distant continental disjunctions in Central Balkans is questionable, since this thermophilous species has been used in medicine for thousands of years and it may have been established post-glacially via anthropogenic mediation. To reveal the origin and the age of its disjunctions, 8 and 11 populations were sampled for molecular and morphometrical investigation, respectively. Sequences of two chloroplast intergenic spacers, 3’rps16-5’trnK and rpl32-trnL, were used for phylogeographic study. Seven haplotypes, non-randomly distributed in space were found as well as high total gene diversity (HT = 0.695) and genetic differentiation (GST = 0.682) showing a phylogeographic structure. According to the strict clock Bayesian analysis, the divergence of an ancient S. officinalis gene pool to two lineages was dated to the Pliocene (3.267 M...a), which coincides with the period when the vast water bodies covered the central part of the Peninsula and presumably divided the paleo-distribution of S. officinalis. A substructured and less diverse continental-Adriatic lineage (CAL, Hd = 0.426, π = 0.00106) and a more diverse, purely Adriatic lineage (PAL, Hd = 0.403, π = 0.00257) do not exhibit signs of recent demographic expansions and currently overlap on the southeastern Adriatic coast, a region designated as the main glacial refugium of S. officinalis. All four populations from the continental Balkans comprising the continental sub-lineage of CAL were almost fixed for a distinct haplotype which was rare in the Adriatic sub-lineage of CAL and absent in PAL.
