Abstract
Iako se transplantacija matičnim ćelijama hematopoeze kao oblik lečenja koristi više decenija, još uvek nije potpuno jasno zašto postoje razlike u brzini oporavka hematopoeze (vremenu prihvatanja kalema, engl. engrafment) kod bolesnika sa istim oboljenjima, kod kojih su primenjeni istovetni mobilizacioni protokoli i režimi kondicioniranja. Za sagledavanje terapeutskog potencijala matičnih ćelija neophodno je poznavanje njihovih funkcionih karakteristika, između ostalog metaboličkog (energetskog) statusa i redoks-spregnutih mehanizama kontrole diferencijacije u različitim ćelijskim stanjima (mirovanje, opredeljivanje, diferencijacija, aktivacija), tokom hematopoeze. Cilj disertacije je da se ispitaju metabolički parametari u matičnim ćelijama hematopoeze periferne krvi bolesnika sa solidnim tumorima (neuroblastom, NB), Juingov sarkom (engl. Ewing Sarcoma, ES) i limfoma (Hočkinov, HL i non Hočkinov, NHL). Ispitivani su ekspresioni profili enzima ključnih metaboličkih puteva energetskog m...etabolizma: glikolize, -oksidacije masnih kiselina, ciklusa limunske kiseline, procesa oksidativne fosforilacije (ND4 subjedinica kompleksa I, Ndufa6 subjedinica kompleksa I, citohrom b, citohrom c i subjedinice II (COX II) i IV(COX IV) citohrom c oksidaze) i same sinteze ATP. Ispitivan je i funkcioni status mitohondrija i nivo mitohondrijalne DNK, ekspresija NADPH oksidaze, ekspresija izoformi azot oksid sintaza i produkcija azot oksida, ekspresija enzima antioksidativne odbrane matičnih ćelija hematopoeze (glutation-peroksidaze, GSH-Px), katalaze, mangan superoksid dismutaze (MnSOD), bakar cink superoksid dismutaze (CuZnSOD) i ekspresija interleukina-6 (IL-6) i faktora nekroze tumora  (engl. tumor necrosis factor , TNF). Eksperimenti su rađeni na uzorcima monojedarnih ćelija periferne krvi pacijenata koji su primali protokol mobilizacije (MP), pacijenata koji nisu primali protokol mobilizacije i od zdravih davalaca (kontrola). Na osnovu dobijenih rezultata zaključuje se da se izdvajaju dva metabolička fenotipa monojedarnih ćelija iz periferne krvi. Prvi fenotip karakterističan je za monojedarne ćelije periferne krvi kod svih ispitivanih tumora, bez primene protokola mobilizacije. Drugi fenotip istovetan je za monojedarne ćelije periferne krvi svih ispitivanih tumora posle primene protokola mobilizacije...

Hematopoietic stem cell transplantation (HSCT) is a standard clinical practice for more than 50 years. However, we still cannot predict with certainty a rate of recovery in the patients with the same disease undergoing identical therapeutic protocols. To determine therapeutic potential of hematopoietic stem cell (HSC), we need to understand better correlation between the metabolic/energy status of the cells and stages of stem cell differentiation during hematopoiesis. Main goal of this work is to investigate metabolic status of peripheral blood mononuclear cells (PBMC) in patients with solid tumors (neuroblastoma, Ewing sarcoma) and lymphoma (Hodgkin and nonHodgkin). We determined expression profiles of key enzymes involved in glycolysis, fatty acid β-oxidation, citric acid cycle, oxidative phosphorylation and adenosine triphosphate (ATP) synthesis: ND4 subunit complexes I, Ndufa6 subunits of complex I, cytochrome b, cytochrome c and subunits II (COX II) and IV (COX IV) cytochrome c ox...idase. Next, we investigated number of mitochondria and their function as well as expression levels of nicotinamide adenine dinucleotide phosphate (NADPH) - oxidase, expression isoforms of nitric oxide synthase and production of nitric oxide, expression of HSC antioxidant enzymes (glutathione-peroxidase (GSH-Px), catalase, manganese superoxide dismutase (MnSOD), copper-zinc superoxide dismutase (CuZnSOD)) and expression of interleukin-6 (IL-6) and tumor necrosis factor  (TNF). Experiments were performed on PBMC samples of patients that were: a) subjected to mobilization of stem cells from bone marrow, b) not subjected to mobilization, and c) healthy donors (control group). Our results suggested that PBMC metabolic phenotypes differ between mobilized and non-mobilized patients, regardless of diagnosis. These two phenotypes represent functional status of the PBSC at the level of anti-oxidative defense, energy metabolism (glycolysis, -oxidation of fatty acids, citric acid cycle, oxidative phosphorylation-OXPHOS), ATP synthesis, mitochondrial DNA (mtDNK), expression of NADPH oxidase and cytokines IL-6 and TNFα. In samples of non-mobilized HL, NHL and NB patients, we detected a strong metabolic activation of PBMC in comparison with control samples from healthy individuals...
