dc.description.abstractUvod. Lupus erythematosus (LE) je pleomorfno multisistemsko oboljenje koje karakteriše slom tolerancije. Proizvodnja autoantitela, deponovanje imunskih kompleksa i aktivacija komplementa dovode do zapaljenja i oštećenja tkiva. S obzirom da su autoantitela specifična za različite nukleusne i citoplazmatske antigene, LE ima širok spektar kliničkih prezentacija, dok tok bolesti karakteriše smenjivanje remisija i egzacerbacija. Danas je široko prihvaćen stav da poremećaj uklanjanja apoptotičnih ćelija i posledična sekundarna nekroza mogu dovesti do razvoja oboljenja koja imaju autoimunsku komponentu, kao što je LE. Iako većina podataka o poremećajima apoptoze kod LE dolazi iz studija na mišjim modelima, postoje i brojne studije kod pacijenata sa sistemskim LE (SLE) koje su pokazale kako pojačanu apoptozu, koja je kod kutanog lupusa indukovana UV zracima, tako i poremećaj uklanjanja apoptotičnih ostataka, u čemu glavnu ulogu igra dezoksiribonukleaza I (DNaza I), glavna nukleaza seruma. Smatra se da je DNaza I jedan od ključnih molekula u razvoju SLE. Postoji nekoliko razloga za ovu tvrdnju: a) dsDNA je predominantan autoantigen kod pacijenata sa SLE, b) DNaza iz bovinog pankreasa uništava antigenost DNK, i c) aktivnost DNaze I u serumu pacijenata sa SLE je često niska i to sniženje je veće kod pacijenata sa aktivnijim SLE. Ciljevi istraživanja. Ispitati da li postoji korelacija između nivoa aktivnosti DNaze I u serumu i indeksa aktivnosti bolesti kod pacijenata sa kutanim LE (CLE) i SLE, kao i imunoseroloških parametara aktivnosti SLE. Utvrditi stepen apoptoze u koži lezije i zdravoj fotoprotektovanoj koži kod pacijenata sa LE i ispitati da li postoji korelacija između stepena apoptoze u koži i nivoa aktivnosti DNaze I u serumu...sr
dc.description.abstractIntroduction. Lupus erythematosus (LE) is a pleomorphic multisystemic disease characterized by the breakdown of tolerance. Production of autoantibodies, immune complex deposition and complement activation leads to inflammation and tissue damage. Given that autoantibodies are specific for different nuclear and cytoplasmic antigens, LE has a wide range of clinical presentations, and course of the disease is characterized by alternating of remissions and exacerbations. Today it is widely accepted that disorders of removal of apoptotic cells and the consequent secondary necrosis can lead to the development of autoimmune diseases, such as the LE. Although most of the data on apoptosis in LE comes from studies in mouse models, there are a number of studies in patients with systemic LE (SLE). These studies showed increased apoptosis, UV-induced in the cutaneous lupus, and disturbance of removal of apoptotic debris, the process in which desoxyribonuclease I (DNase I), the principal serum nuclease, plays the major role. DNase I is thought to be one of the key molecules in the development of SLE for several reasons: a) double-stranded DNA (dsDNA) is the predominant autoantigen in patients with SLE, b) DNase from bovine pancreas destroys the antigenicity of DNA, and c) the DNase I activity in sera of SLE patients is often low and that decrease is more profound in patients with active SLE. Aims of the investigation. To examine whether there is a correlation between the level of serum DNase I activity and indices of the disease activity in patients with cutaneous LE (CLE) and SLE, and immunological parameters of SLE activity.
