Abstract
Poznato je da efekti interakcije biljka - Trichoderma zavise od genotipa biljke i soja Trichoderma, međutim molekularni mehanizmi ovog fenomena jos uvek nisu razjašnjeni. Glavni cilj ove doktorske disertacije je definisanje molekularno-fizioloških pokazatelja pozitivnog odgovora biljaka na primenu gljiva roda Trichoderma. U cilju pronalaženja ovih odgovora vršene su analize ekspresije gena, biohemijske i morfo-fiziološke analize. Gljive roda Trichoderma stimulišu klijanje semena paradajza, pri čemu značajno utiču na promene koje se odigravaju na nivou proteina kao i pektina i/ili ksiloglukana u radikulama semena. U okviru brzih reakcija biljaka na prisustvo T. brevicompactum najznačajnije su bolje adaptivne osobine na prestanak zalivanja, odnosno veća provodljivost stoma i manji porast ABA, što pozitivno utiče na brzinu fotosinteze, a samim tim i na rast biljaka. Dugoročni efekti primene T. harzianum i T. brevicompactum su različiti kod ispitivanih sorti paradajza. T. harzianum stimuli...še provodljivost stoma i intracelularnu koncentraciju CO2 optimalno zalivanih biljaka paradajza, dok T. brevicompactum stimuliše nefotohemijsko gašenje fluorescencije hlorofila i trenutnu efikasnost korišćenja vode. T. harzianum kod biljaka ublažava efekte suše smanjenjem temperature lista i inhibicije fotosinteze. Utvrđeno je da T. harzianum značajno ubrzava formiranje plodova, dok ujedno povećava sadržaj flavonoida u plodu čime doprinosi njegovom kvalitetu. Analize ekspresije gena (TPX1, PINII, PR1b1, SlJAF13, FLS, NCED1 i C3H) i kvalitativne analize fenolnih jedinjenja ukazuju na međuzavisnost i kompleksnost signalnih puteva i sekundarnih metabolita. Utvrđeno je da je povišena ekspresija svolenina povezana sa uspešnijom kolonizacijom korena paradajza od strane gljiva roda Trichoderma.

The effects of plant - Trichoderma interactions are depended on the plant genotype and the Trichoderma strain. However, the molecular mechanisms of this phenomenon have not yet been elucidated. The main aim of this PhD dissertation is to define molecular-physiological indicators of the plants positive response to the application of fungi of the genus Trichoderma. In order to find these answers, gene expression analyses, biochemical and morpho-physiological analyses were performed. The fungi of the genus Trichoderma stimulate the germination of tomato seeds, significantly influencing the changes that take place at the level of proteins as well as pectin and/or xyloglucan in the seed radicals. Within the fast responses of plants to the application of T. brevicompactum, the most important effects were better adaptive characteristics to the cessation of watering, i.e., higher stomatal conductance and a lower level of ABA, which positively affect the rate of photosynthesis as well as plant ...growth. The long-term effects of T. harzianum and T. brevicompactum showed different plant answers for the examined tomato varieties. T. harzianum stimulates stomatal conductance and intracellular CO2 concentration of optimally watered tomato plants, while T. brevicompactum stimulates nonphotochemical chlorophyll fluorescence quenching and water use efficiency. T. harzianum in plants mitigates the impacts of drought by reducing leaf temperature and inhibiting photosynthesis. The results showed that T. harzianum significantly stimulates fruit formation, while also increasing the content of flavonoids in the fruit, which contributes to its quality. Gene expression analyses (TPX1, PINII, PR1b1, SlJAF13, FLS, NCED1 and C3H) and qualitative analyses of phenolic compounds indicate the interdependence and complexity of signaling pathways and secondary metabolites. Better colonization of plant roots by Trichoderma is determined by higher expression of the swolenin gene in tomato roots.
