Cilj ovog istraživanja je da ispita uticaj akutnog oštećenja bubrega izazvanog
nefrotoksičnim agesima: gentamicinom, diklofenom i folnom kiselinom na
funkciju miokarda i inflamacijsko-oksidacioni status pacova. Istraživanje je
obuhvatilo 100 pacova Wistar albino soja muškog pola starosti 6 nedelja, prosečne
telesne mase oko 200 grama. Pacovi su podeljeni u tri eksperimentalne grupe od kojih
je svaka tretirana različitim nefrotoksičnim agensom: diklofenom, gentamicinom i
folnom kiselinom i kontrolnu koja je tretirana fiziološkim rastvorom. Nakon
eksperimentalnog tretmana i žrtvovanja sprovedena su ispitivanja na izolovanom
srcu metodom po Langendorff-u praćenjem parametara srčane funkcije: dp/dt max, dp/dt
min, SLVP, DLVP, HR i CF, dok su spektrofotometrijskim metodama u efluentu i
plazmi određeni parametri oksidacionog stresa (TBARS, NO2

, O2
-
, H2O2) i u lizatu
eritrocita parametri antioksidacione zaštite (CAT, SOD, GSH). ELISA metodom u
plazmi određeni su biomarkeri in...flamacije (TNF-α, IL-1, IL-6). Rezultati ove studije
pokazuju da akutno oštećenje bubrega utiče na kardijalnu hemodinamiku, sistemska i
kardijalna oksidacijska svojstva kao i na sistemsku inflamaciju i pomaže u
rasvetljavanju efekata akutnog oštećenja bubrega na funkciju i redoks potencijal
kardiovaskularnog sistema.
The aim of this study was to examine the effect of acute kidney injury caused by
nephrotoxic ages: gentamicin, diclofen and folic acid on myocardial function and the
inflammatory-oxidative status of rats. The study included 100 ѕix week old male Wistar
albino rats, with an average body weight about 200 grams. Rats were divided into three
experimental groups, each of them were treated with a different nephrotoxic agent:
diclofenac, gentamicin and folic acid, and a control group treated with saline. After
experimental treatment and sacrifice, studies on the isolated heart were performed by the
Langendorff method by monitoring the parameters of cardiac function: dp / dt max, dp / dt
min, SLVP, DLVP, HR and CF. The parametres of oxidative stress (TBARS, NO2-, O2-,
H2O2) in effluent and plasma were determined by spectrophotometric methods as well as
parameters of antioxidant protection (CAT, SOD, GSH) in erythrocyte lysate. Biomarkers of
inflammation in plasma (TNF-α, IL-1, IL-6)... were determined by ELISA method. The results
of this study show that acute renal impairment affects cardiac hemodynamics, systemic and
cardiac oxidative properties as well as systemic inflammation and helps to elucidate the
effects of acute renal impairment on the function and redox potential of cardiovascular
system.
