dc.description.abstractCilj ove disertacije bio je ispitivanje antitumorskog potencijala novosintetisanih rutenijum(II)-arenskih kompleksa sa bioaktivnim ligandima in vitro. Ispitivani kompleksi sadrže ligande: 1) indometacin (L1) i mefenaminsku kiselinu (L2), kao predstavnike nesteroidnih antiinflamatornih lekova (kompleksi K1-K4); 2) polipiridin (L3) (kompleksi K5-K7); i 3) 2-amino-4-metilbenzamid (L4) i 3-amino-N-metilbenzamid (L5), kao analoge inhibitora poli(ADP-ribozil) polimeraze (PARP) 3-aminobenzamida (3-AB) (kompleksi K8-K11). Antiproliferativni potencijal ovih kompleksa je ispitan primenom 3-(4,5-dimetiltiazol-2-il)-2,5-difeniltetrazolijum bromid (MTT) testa na humanim tumorskim ćelijskim linijama i višećelijskim tumorskim sferoidima, u poređenju sa cisplatinom. Mehanizmi dejstva najaktivnijih kompleksa su ispitani na ćelijskoj liniji koja je pokazala najveću senzitivnost, upotrebom protočne citofluorimetrije, fluorescentne mikroskopije i masene spektrometrije. Rezultati su pokazali da L1 doprinosi aktivnosti i selektivnosti kompleksa K1 i K3 za ćelije trostruko-negativnog karcinoma dojke MDA-MB-231. Kompleksi K1 i K3 dovode do zaustavljanja MDA-MB-231 ćelija u S fazi ćelijskog ciklusa i apoptoze. Cimen u strukturi kompleksa K7 doprinosi selektivnosti za ćelije malignog melanoma A375, ali u odnosu na cisplatinu, K7 pokazuje nižu unutarćelijsku akumulaciju, odsustvo citotoksičnosti i ograničenu aktivnost na sferoidima. Iako su se kompleksi K9 i K11 (sa cimenom) pokazali kao potentniji PARP-1 inhibitori u odnosu na kompleks K8 (sa toluenom) i 3-AB, K8 pokazuje najveći antiproliferativni potencijal i afinitet prema nukleusu i DNK ćelija BRCA-mutiranog trostruko-negativnog karcinoma dojke HCC1937. Dobijeni rezultati pružaju osnov za dalje istraživanje mehanizama dejstva ovakvog tipa kombinovanih molekula u svrhu razvoja novih potencijalnih antitumorskih lekova.sr
dc.description.abstractThe aim of this thesis was to investigate the antitumor potential of newly synthesized ruthenium(II)-arene complexes with bioactive ligands in vitro. Investigated complexes contain ligands: 1) indomethacin (L1) and mefenamic acid (L2), as representatives of nonsteroidal antiinflammatory drugs (complexes K1-K4); 2) polypyridine (L3) (complexes K5-K7); and 3) 2-amino-4-methylbenzamide (L4) and 3-amino-N-methylbenzamide (L5), as analogs of poly(ADP-ribosyl) polymerase (PARP) inhibitor 3-aminobenzamide (complexes K8-K11). The antiproliferative potential of these complexes was examined on human tumor cell lines and multicellular tumor spheroids, in comparison to cisplatin, by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) test. The mechanisms of action of the most active complexes were examined on the most sensitive cell line by flow cytofluorimetry, fluorescence microscopy and mass spectrometry. The results showed that L1 contributes to the activity and selectivity of the K1 and K3 complexes towards triple-negative breast cancer cells MDA-MB-231. The K1 and K3 complexes cause cell cycle arrest in the S phase and apoptosis of MDA-MB-231 cells. Cymene in the structure of the K7 complex contributes to selectivity for A375 malignant melanoma cells, but compared to cisplatin, K7 showed a lower intracellular accumulation, absence of cytotoxicity and limited activity on spheroids. Although the K9 and K11 complexes (with cymene) have shown to be more potent PARP-1 inhibitors than the K8 complex (with toluene) and 3-AB, K8 showed the greatest antiproliferative potential and affinity for the nucleus and DNA of BRCA-mutated triple-negative breast cancer cells HCC1937. The obtained results provide a basis for further research into the mechanisms of action of this type of combined molecules for the purpose of development of novel potential antitumor drugs.en
