dc.descriptionThis doctoral dissertation is a set of studies aimed at obtaining relevant data on the content of active compounds and the pharmacological effects of the clary sage extract, Salvia sclarea L. In traditional medicine, S. sclarea is known as an aromatic and medicinal plant species most commonly used in various digestive, respiratory and menstrual disorders, inflammatory processes, cough, gingivitis, aphthae, stomatitis, etc. Its healing properties have not been fully investigated thus far, so these results will provide a significant contribution to modern phytotherapy. The above-ground parts of clary sage, used as plant material, were collected in the period 2011-2014 in Malča, near Niš. The collected plant material was subjected to morphological-anatomical characterization and extraction. Ten extracts were prepared from each year by the extraction with various concentrations of ethanol and methanol using maceration, ultrasound and digestion techniques. The morphological and anatomical analysis of the above-gound organs of S. sclarea and the elements of the drug (Salviae sclareae herba) indicates the presence of two functional types of trichomes: non-glandular and glandular. Unicellular and multicellular glandular trichomes, as well as capitate and peltate glandular trichomes, with a larger number of subtypes are present. Chemical investigations determined significant amounts of total phenols, tannins, flavonoids and hydroxycinnamic acid derivatives in the extracts. HPLC analysis identified and quantified seven compounds in all extracts: rosmarinic and caffeic acid, apigenin, luteolin, salvigenin, apigenin- and luteolin-7-O-glucoside. The dominant compound in all the extracts was rosmarinic acid. Pharmacological studies involved the definition of antioxidant, antimicrobial, anti-inflammatory and spasmolytic effects of the extracts. All clary sage extracts showed significant antiradical, especially antilipoperoxidant, potentials. The extracts influenced the growth of Gram (+) bacteria more effectively than Gram (-) and were better bacteriostatic than bactericidal agents. Especially important is the good effect on Staphylococcus aureus. An experimentally (lipopolysaccharide) induced periodontitis on rats showed a significant anti-inflammatory effect of the ethanol extract, prepared with 80% ethanol by ultrasound extraction. The investigated extracts exhibited significant relaxant effects on spontaneous and induced contractions of smooth muscles of the isolated ileum and trachea of the rats. The chemical characterization of S. sclarea extracts showed that the differences in their composition reflected the manifestation of pharmacological activities. Further research into clary sage extracts should be directed towards in vivo and clinical studies so as to examine their potential application as effective medicinal preparations.en
