dc.description.abstract | Kako su višedecenijska istraživanja pokazala kardioprotektivna dejstva antihipertenzivnih lekova, zainteresovalo nas je da istražimo njihov efekat na ishemijsko-reperfuzionu (I/R) povredu srca. Cilj studije je bio da se utvrde efekti subhronične terapije različitim antihipertenzivnim lekovima na I/R disfunkciju srca kod hipertenzivnih pacova po modelu globalne ishemije na izolovanom srcu pacova. Istraživanje je sprovedeno na 80 spontano hipertenzivnih Wistar Kyoto pacova mužjaka, grupisanih u 8 grupa. Životinje su tretirane četiri nedelje sa 10 mg/kg hidrohlorotiazida, indapamida, zofenoprila, spironolaktona ili njihovih kombinacija (ZHS, ZIS) per os. Nakon tretmana sprovedena su hemodinamska merenja, ehokardiografija i procena funkcije miokarda prema Langendorff retrogradnoj metodi perfuzije. Indukovani model globalne ishemije je podrazumevao primenu 20 minuta ishemije, a zatim 30 minuta reperfuzije na srce (I20:R30). Markeri oksidativnog stresa određivani su iz koronarnog venskog efluenta, plazme i lizata eritrocita spektrofotometrijski. Uzorci tkiva srca i bubrega su patohistološki analizirani. Tretman kombinacijom ACE-inhibitora i duretika značajno je snizio krvni pritisak spontanohipertenzivnih pacova, ublažio hipertrofiju leve komore i povećao ejekcionu frakciju, dok na oksidacioni status nije delovao. S druge strane samostalna primene sva tri ispitivana diuretika je pokazala izuzetan antioksidativni potencijal. Patohistološka analiza uzoraka tkiva srca i bubrega, ukazuje da subhronična primena antihipertenzivnih agenasa ne dovodi do značajnih promena u ovim organima. Praćenje srčane funkcije spontanohipertenzivnih pacova ex vivo dalo je uvid u kardioprotektivne efekte indapamida. | sr |
dc.description.abstract | Аntihypertensive drugs have positive benefits in cardiovascular disease, so we sought to examine their effect on ischaemia-reperfusion (I/R) injury of heart. The study's objective was to ascertain results of subchronic therapy of аntihypertensive agents on I/R dysfunction of the heart in spontaneously hypertensive rats (SHR) by a model of global ischemia on isolated heart of rat. The research included 80 Wistar Kyoto SHR, male sex, grouped into 8 groups. The animals were treated 4 weeks (10 mg/kg) with hydrochlorothiazide, indapamide, spironolactone, zofenopril or their combinations (ZHS, ZIS) per os. After treatment we analysed hemodynamic measurements, echocardiography, and myocardial function according to the Langendorff retrograde perfusion method. An induced global ischemia model involved applying 20 minutes of ischemia followed by 30 minutes of reperfusion to the heart. Oxidative stress markers were determined spectrophotometrically from coronary venous effluent, plasma and erythrocyte lysate. Heart and kidney tissue samples were pathohistologically analyzed. Treatment with ZHS and ZIS lowered the blood pressure of SHR, alleviated left ventricular hypertrophy and increased the ejection fraction, while it had no effect on the oxidation status. On the other hand, all three tested diuretics showed exceptional antioxidant potential. Pathohistological analysis indicates that subchronic administration of antihypertensive agents does not lead to significant changes in heart and kidney. Cardiac function monitoring of SHR provided insight into cardioprotective effects of indapamide. | en |
