Ispitivanje citotoksičnosti kompleksa paladijuma(II) sa derivatima tiazola kao ligandima na ćelijama tumora prostate in vitro

Biološki aktivne supstance koje u svojoj strukturi sadrže metale su budućnost
antitumorske terapije. Kompleksi metala u zavisnosti od svoje strukture i hemijskih
osobina pokazuju manji ili veći stepen antitumorskog efekta na različite ispitivane
ćelijske linije.
Supstance koje u svojoj strukturi sadrže metal paladijum su pokazale veoma jako
antitumorsko dejstvo na cisplatin-rezistentne tumorske ćelijske linije. Svrha,
odnosno cilj naše studije je bio da detaljno testiramo biološki efekat pet Pd(II)
kompleksa (obeleženih kao C1-C5) i odgovarajućih liganada (obeleženih kao L1-L5), na
tumorske ćelije humanog karcinoma prostate PC-3 i DU-145 kao i na ćelijskoj liniji
maligno neizmenjenih fibroblasta MRC-5.
Kao metode za određivanje biološke aktivnosti Pd(II) kompleksa i
odgovarajućih liganada koristili smo MTT test, Annexin V/PI test, analizu ćelijskog
ciklusa primenom PI bojenja i analizu ključnih proteina apoptoze.
Obradom i detaljnom analizom dobijenih rezlutata pokazano je da... svi testirani
Pd(II) kompleksi demonstriraju veoma jak i antitumorski efekat na PC-3 i DU-145
ćelijske linije in vitro. Takođe, prethodno pomenuti kompleksi su ispoljili veoma slab
antitumorski efekat na maligno neizmenjene fibroblaste MRC-5, čime je jasno
pokazana visoka selektivnost testiranih supstanci. Ispitivani ligandi L1-L5 su
pokazali veoma slab antitumorski efekat na svim testiranim ćelijskim linijama, čime
je jasno dokazana ključna uloga paladijuma.
Dalje, testirani, Pd(II) kompleksi smanjuju vijabilnost PC-3 i DU-145 na dva
načina, indukovanjem procesa apoptoze ili zastojem u različitim fazama ćelijskog
ciklusa. Preciznije, ispitivani Pd(II) kompleksi indukuju unutrašnji
mitohondrijalni put, povećavajući relatisni odnos Bax/Bcl-2, a zatim indukujući
aktivaciju kaspaze-3.
Biologically active substances that contain metals in their structure are the future of antitumor
therapy. Depending on their structure and chemical properties, metal complexes show a lower
or higher degree of antitumor effect on the different tested cell lines.
Substances that contain palladium metal in their structure have shown a very strong antitumor
effect on cisplatin-resistant tumor cell lines. Our study aimed to test in detail the biological
effect of five Pd(II) complexes (marked as C1-C5) and the corresponding ligands (marked as
L1-L5) on PC-3 and DU-145 human prostate cancer tumor cells. as well as on the MRC-5
fibroblast cell line without malignant changes.
As methods for determining the biological activities of Pd(II) complexes and corresponding
ligands, we used the MTT test, Annexin V/PI test, cell cycle analysis using PI staining, and
analysis of key apoptotic proteins.
Results showed that all tested Pd(II) complexes demonstrated a very strong antitumor effect on ...
PC-3 and DU-145 cell lines in vitro. Also, the previously mentioned complexes exhibited
insignificant antitumor effects on non-cancerous MRC-5 fibroblasts, which demonstrated the
high selectivity of the tested substances. The tested ligands L1-L5 showed an insignificant
antitumor effect on all tested cell lines, thus clearly proving the key role of palladium.
Furthermore, the tested Pd(II) complexes reduce the viability of PC-3 and DU-145 in two ways,
by inducing the process of apoptosis or by keeping them in different phases of the cell cycle.
More precisely, the tested Pd(II) complexes induce the inner mitochondrial pathway of
apoptosis, increasing the relative ratio of Bax/Bcl-2 proteins, and then inducing the activation
of caspase-3.
