Morphological variation and developmental anomalies in Lepidurus couesii Packard, 1875 (Branchiopoda, Notostraca) from Serbia

dc.description.abstractU disertaciji je ispitano nekoliko aspekata biologije vrste Lepidurus couesii iz Srbije. Prvobitno je vrsta identifikovana analizom morfoloških karaktera koji su u dosadašnjoj literaturi definisani kao važni taksonomski karakteri. Zatim je ispitana histološka građa gonada kako bi se jedinke kategorisale kao ženke, mužjaci ili hermafroditi, kao i radi ispitivanja mogućih načina reprodukcije populacija iz Srbije. Morfološka varijabilnost vrste je dodatno opisana analizom variranja oblika dve morfološke celine – telzona sa kaudalnom laminom i karapaksa. Na kraju, ispitane su i definisane anomalije koje su nastale tokom razvića jedinki, kao i prisutne kvalitativne promene morfoloških struktura. Istraživanjem su obuhvaćene populacije račića koje nastanjuju efemerna vodena staništa. Populacije roda Lepidurus su uzorkovane sa dva lokaliteta na području Srema, u okolini reke Save. U prvom delu studije izvršena je morfometrijska analiza kontinuiranih karaktera i kvantifikacija merističkih dijagnostičkih karaktera. Kvalitativni karakteri su predstavljeni odnosima određenih morfometrijskih karaktera. Dodatni kvalitativni morfološki karakteri su analizirani opisno. Rezultati su upoređeni sa relevantnim literaturnim podacima o morfološkim karakteristikama grupe Notostraca. Pol jedinki i osnovne odlike gametogeneze su ispitane primenom metoda svetlosne i elektronske mikroskopije. Morfološko variranje telzona sa kaudalnom laminom i karapaksa je ispitano i primenom metoda geometrijske morfometrije. Ovakav pristup je omogućio geometrijsku analizu oblika navedenih struktura, koncept koji do sada nije primenjen u okviru roda Lepidurus. Takođe, ispitani su obrasci alometrijske i nealometrijske komponente variranja oblika dve morfološke strukture. Kako bi se definisali i analizirali atipični fenotipovi, jedinke su detaljno pregledane pomoću binokularnog stereomikroskopa. Sve uočene morfološke promene su zabeležene. Rezultati su pokazali da se morfološke odlike jedinki iz analiziranih populacija najviše podudaraju sa karakteristikama vrste Lepidurus couesii. Histološke analize gonada nisu pokazale prisustvo tkiva koje odgovara ovotestisima hermafroditnih jedinki. U skladu sa dobijenim rezultatima, jedinke su determinisane kao ženke ili mužjaci u zavisnosti od prisustva, odnosno odsustva, jajnih kesa na jedanaestom paru torakopoda. Ženke su daleko brojnije od mužjaka – od 219 prikupljenih jedinki, pronađeno je samo devet mužjaka. Analize metodama geometrijske morfometrije na uzorku ženki su pokazale da je veliki deo variranja oblika telzona sa kaudalnom laminom opisan alometrijskim promenama oblika (više od 70%). Alometrija objašnjava znatno manji deo variranja oblika karapaksa (oko 5%). Preliminarni rezultati ukazuju da je prisutan polni dimorfizam u obliku navedenih morfoloških struktura. Ženke u proseku imaju širi telzon i kraću kaudalnu laminu u odnosu na mužjake, kao i duži i suženiji karapaks. Ustanovljene razvojne anomalije prisutne u populacijama su klasifikovane na malformacije i disrupcije. Malformacije uključuju nepotpuno razvijene i spiralno raspoređene segmente. Takođe su registrovane i asimetrično oblikovane kaudalne lamine. Disrupcije uglavnom obuhvataju ozlede ekstremiteta, karapaksa, telzona sa kaudalnom laminom, kao i kvalitativne promene koje ukazuju na parazitsku infekciju. Na osnovu dobijenih rezultata, smatra se da se populacije vrste L. couesii iz Srbije razmnožavaju prvenstveno partenogenezom. dc.description.abstractWithin this dissertation, several aspects of the biology of Lepidurus couesii, species inhabiting Serbia, were investigated. Initially, an analysis of morphological characters, defined as having taxonomic importance in the previous literature, was performed to identify the species. Then, the histological structure of the gonads was examined to categorize the individuals as females, males, or hermaphrodites, as well as to investigate possible ways of reproduction of populations from Serbia. Morphological variability in the species was also studied by analyzing shape variation in two morphological structures – the telson with a caudal lamina and the carapace. Finally, the anomalies that occurred during the development of individuals were examined and defined, along with the qualitative changes in the morphological structures present. The research included populations of small crustaceans inhabiting temporary aquatic habitats. Populations of Lepidurus were sampled from two localities in the area of Srem, in the vicinity of the Sava river. At the beginning of the study, morphometric measurements and meristic counts of diagnostic characters were performed. Qualitative characters were represented by ratios of certain morphometric characters. Additional qualitative morphological characters were analyzed descriptively. The results were compared with relevant literature data on the morphological characteristics of notostracans. The sex of specimens and general features of gametogenesis were characterized using light and electron microscopy. Morphological variation in the telson with the caudal lamina and the carapace was examined using geometric morphometrics methods. This approach enabled geometric shape analysis, a concept that has not been applied so far within the genus Lepidurus. The patterns of allometric and allometry-free shape variation in two morphological structures were examined. Individuals were also thoroughly investigated with a binocular stereomicroscope to define and analyze atypical phenotypes. Observed morphological changes were recorded. The results showed that the morphological features of the individuals from the analyzed populations most closely coincide with the characteristics of the Lepidurus couesii species. Histological analyses of the gonads did not show the presence of tissue found in the ovotestes of hermaphrodite individuals. Accordingly, the individuals were determined as females or males depending on the presence or absence of egg sacs on the eleventh pair of thoracopods. Females are far more numerous than males – out of 219 collected individuals, only nine males were found. Geometric morphometric analyses conducted on females showed that a large proportion of telson with the caudal lamina shape variation is described by size (more than 70%). Allometry explains a much smaller proportion of carapace shape variation (about 5%). Preliminary results indicate the occurrence of sexual shape dimorphism in the mentioned morphological structures. Females typically have a wider telson and a shorter caudal lamina than males, along with a longer and narrower carapace. Developmental anomalies present in populations are classified as malformations and disruptions. Among identified malformations were incompletely developed and spirally arranged body rings.
