dc.description.abstractCilj ove studije bio je da ispita i uporedi efekate prekondicioniranja farmakološkim agensima, hiperbaričnom oksigenacijom, kao i njihove kombinovane primene na animalnom modelu. U studiju je bilo uključeno 96 pacova (Wistar albino soj, muški pol, starosti 8 nedelja, telesne mase 200±50g). U prvi deo studije (in vivo) bilo je uključeno 48 životinja koje su bile izložene prekondicioniranju hiperbaričnom oksigenacijom (HBO) u trajanju od 14 dana. Nakon toga je usledio drugi deo studije (ex vivo) koji je obuhvatio sve životinje koje su bile uključene u studiju. Izolovana srca pacova su perfundovana metodom retrogradne perfuzije po Langendorfu pri konstantnom koronarnom perfuzionom pritisku od 70 sm H2O, a nakon stabilizacije srca su podeljena u dve kontrolne (kontrola bez prekondicioniranja i HBO – srca pacova koji su bili izloženi hiperbaričnoj oksigenaciji) i 6 eksperimentalnih grupa koje su se međusobno razlikovale u primenjenom manevru prekondicioniranja (verapamil, amlodipin, nikorandil ili kombinovano HBO + verapamil, HBO + amlodipin, HBO + nikorandil). Potom su srca bila izlagana globalnoj ishemiji u trajanju od 20 minuta i reperfuziji u trajanju od 30 minuta. Tokom čitavog eksperimentalnog protokola beleženi su kardiodinamski parametri, koronarni protok i sakupljan je koronarni venski efluent iz koga su određivani parametri oksidacionog stresa. Dobijeni rezultati jasno ukazuju da blokiranje kalcijumskog kanala ili aktivacija ATR-zavisnih kalijumskih kanala može posredovati zaštitnom efektu predkondicioniranja miokarda, svi ispitivani lekovi imaju korisna kardioprotektivna svojstva povezana sa nižim vrednostima glavnih prooksidacionih molekula.sr
dc.description.abstractТhe aim of this study was to examine and compare the effects of preconditioning with pharmacological agents, hyperbaric oxygenation, and their combined applications on the animal model. The study included 96 rats (Wistar albino, male gender, 8 weeks old, body weight 200 ± 50g). In the first part of the study (in vivo), 48 animals were exposed to hyperbaric oxygenation (HBO) over 14 days. A second part of the study (ex vivo) included all the animals involved in the study. Isolated heart rats were perfused by the Langendorff retrograde method at a constant coronary pressure of 70 cm H2O, and after stabilization period heart were divided into two controls (control without preconditioning and HBO - heart of rats exposed to hyperbaric oxygenation) and 6 experimental groups differed in the applied preconditioning maneuver (verapamil, amlodipine, nicorandil or combined HBO + verapamil, HBO + amlodipine, HBO + nicorandil). Then, the heart was exposed to global ischemia for 20 minutes and reperfusion for 30 minutes. During the entire experimental protocol, cardiodynamic parameters and coronary flow were recorded, while a coronary venous effluent was collected form determination of oxidative stress parameters. Obtained results clearly indicate that blocking of the calcium channel or activation of ATP-sensitive K+ channel can mediate the protective effect of myocardial preconditioning; all investigated drugs have useful cardioprotective properties associated with lower values of the major prooxidation molecules.en
