Cilj ove studije je bio da se odredi uticaj ekspresije receptora za estrogene i
progesteron na angiogenezu, proliferaciju i apoptozu ćelija mioma kod žena u pre i
postmenopauzi. Ovo je bila studija preseka, kliničko-eksperimentalna, retrospektivna,
neintervencijska studija u polju istraživanja fundamentalnih mehanizama patogeneze
bolesti, korišćenjem patohistološkog materijala iz postojeće arhive. Istraživanjem je
obuhvaćeno 76 pacijentkinja sa dijagnostikovanim lejomioima uterusa, operativno
lečenim na Klinici za ginekologiju i akušerstvo, Kliničkog centra Kragujevac, Srbija.
Prema menstrualnom statusu, formirane su dve eksperimentalne podgrupe. Prva grupa
bile su žene u premenopauzi (PreM) (n = 35; 46,2 ± 5,02 godina), a druga grupa bile su
žene u postmenopauzi (PostM) (n = 41; 60,25 ± 5,41 godina). Korišćen je metod
standardnog H&E bojenja za isečke mioma, miometrijuma i endometrijuma, kao i
imunohistohemijski metod za određivanje ekspresije ERα, ERβ, PRa, VEGF, CD105, Ki...-
68 i Caspase -3. Ekspresija receptora za progesteron je bila više izražena u miomima i
miometrijumu kod premenopauznih žena u poređenju sa miomima i miometrijumom kod
žena u postmenopauzi. Ekspresija Caspase-3 je statistički značajno veća u grupi PostM
žena u poređenju sa PreM grupom. Ekspresija ERα i ERβ nije bila značajno različita
imeđu grupa ni u uzorcima mioma ni miometrijuma. Prema našim podacima, ekspresija
receptora za progesteron je imala veći uticaj na apoptozu i proliferaciju ćelija nego
ekspresija receptora za estrogene. Povećana ekspresija receptora za progesteron kod
žena u PreM u miomu i miometrijumu, verovatno je dovela do smanjenja ekspresije
apoptotskog markera kod PreM žena.
The aim of this study was to determine the effects of the estrogens and progesterone receptors
status on angiogenesis, proliferation and apoptosis of myoma cells in premenopausal and
postmenopausal women. This was a cross section, clinical-experimental, retrospective, noninterventional study in the field of the study of fundamental pathogenesis mechanisms of disease
using pathohistological materials from the existing archive. The research included 76 patients
diagnosed with uterine leiomyomas, operatively treated in the Clinic for Gynecology and
Obstetrics, Clinical Centre Kragujevac, Serbia. According to the menstrual status, two
experimental subgroups were formed. The first group was premenopausal women (PreM) (n=35;
46.2±5.02 year old), and the second group was postmenopausal women (PostM) (n=41;
60.25±5.41 year old). H&E staining for myoma and myometrium was conducted as well as
immunohistochemistry for ERα, ERβ, PRа, VEGF, CD105, Ki678 and Cas-3. Progesterone
receptor wa...s over expressed in myoma and myometrium of premenopausal compared to myoma
and myometrium of postmenopausal women. Expression of Caspase 3 was statistically
significant increased in PostM women compared to PreM group. ERα and ERβ were not changed
among groups neither in myoma nor in myometrium samples. According to our data, PRа had
higher influence on apoptosis and cell growth than estrogen receptors. Since PRа was increased
in PreM in both myoma and myometrium, probably this expression led further to lower
expression of apoptotic marker in PreM women.
